Overview

TERN-201 is a highly selective, irreversible, covalent small molecule inhibitor of semicarbazide-sensitive amine oxidase (SSAO), also known as vascular adhesion protein-1 (VAP-1). It is being developed primarily for the treatment of non-alcoholic steatohepatitis (NASH). SSAO/VAP-1 is a transmembrane protein highly expressed in hepatic vascular endothelial cells and plays a role in the recruitment and transmigration of white blood cells into the liver, contributing to inflammation and fibrosis. By inhibiting SSAO/VAP-1, TERN-201 reduces oxidative stress and inflammatory cell infiltration in the liver. Clinical studies have shown that oral administration of TERN-201 results in potent, sustained inhibition of plasma SSAO activity with good tolerability and no significant safety concerns observed. The drug has received Fast Track designation from the FDA for NASH[1][5][6][7].
