Overview

TERN-501 is an orally administered, small molecule thyroid hormone receptor beta (THR-β) agonist developed for the treatment of nonalcoholic steatohepatitis (NASH). It is highly selective for thyroid hormone receptor beta over thyroid hormone receptor alpha, with a 23-fold greater selectivity, which minimizes off-target effects such as cardiotoxicity. TERN-501 demonstrates enhanced liver distribution and high metabolic stability. In clinical trials, it has shown significant reductions in liver fat content and improvements in markers of liver fibro-inflammation and lipid metabolism. The drug has also been studied in combination with other agents such as FXR agonists to further improve efficacy without increasing LDL cholesterol levels. TERN-501 received Fast Track designation from the FDA for NASH[4][5][7][8].
