TERN-611 is an orally bioavailable, small molecule antagonist of the glucose-dependent insulinotropic polypeptide receptor (GIPR). Developed by Terns Pharmaceuticals (and subsequently part of Merck's portfolio following acquisition), it is currently in early-phase clinical development for the treatment of obesity and other metabolic disorders. The therapeutic rationale for TERN-611 is based on human genetic data suggesting that reduced GIPR signaling is associated with lower body mass index (BMI) and protection against obesity. By antagonizing the GIPR, TERN-611 aims to provide a potent, weight-loss-inducing therapy that can be administered as a convenient oral alternative to injectable incretin-based treatments. Preclinical studies have demonstrated that TERN-611 is highly selective for GIPR and exhibits favorable pharmacokinetic properties for once-daily oral dosing.
