Overview

Teropavimab is a broadly neutralizing monoclonal antibody (bnAb) under clinical investigation for the treatment of HIV-1 infection. It is derived from the antibody 3BNC117 and has been engineered with LS mutations to extend its half-life in the body. Teropavimab specifically targets the CD4-binding site of the HIV-1 envelope glycoprotein gp120, thereby blocking viral entry into host CD4+ cells—an essential step in HIV infection. The drug is being developed as part of long-acting combination regimens with other bnAbs (such as zinlirvimab) and capsid inhibitors (such as lenacapavir), aiming to provide durable virologic suppression with infrequent dosing. Clinical trials have shown that this approach can maintain viral suppression for at least six months in people living with HIV who are highly sensitive to these antibodies[1][5][6][7].
