TES-ONC-07 (also known as TES-4207) is an orally available, first-in-class small molecule antagonist of the orphan nuclear receptor NR2F6 (nuclear receptor subfamily 2 group F member 6), developed by TES Pharma for cancer immunotherapy. NR2F6 acts as an intracellular immune checkpoint that is upregulated in tumor-infiltrating lymphocytes (TILs) and transcriptionally represses key cytokines such as interleukin-2 (IL-2) and interferon-gamma (IFNγ), thereby limiting immune cell activation and infiltration in the tumor microenvironment. By binding to NR2F6, TES-ONC-07 induces its detachment from target gene promoters, restoring cytokine production and enhancing anti-tumor T-cell responses. In preclinical models of melanoma and ovarian cancer, TES-ONC-07 has demonstrated the ability to convert immunologically cold tumors into hot tumors, significantly reducing tumor growth both as a monotherapy and in combination with anti-PD-1 therapy.
