Overview

Teserpaturev (G47Δ) is a third-generation, triple-mutated recombinant oncolytic herpes simplex virus type 1 (HSV-1) developed by Daiichi Sankyo. It is engineered with three specific mutations: deletions in both copies of the γ34.5 gene, an inactivation of the ICP6 (UL39) gene, and a deletion of the α47 gene along with the overlapping US11 promoter. These modifications collectively enhance tumor-specific replication, improve stimulation of antitumor immunity through increased MHC class I presentation, and maintain a high safety profile by restricting replication to cancer cells. In June 2021, it received conditional and time-limited approval in Japan under the brand name Delytact for the treatment of residual or recurrent malignant glioma, making it the first approved oncolytic virus therapy for brain tumors in the world. It is administered via repeated intratumoral (stereotactic) injections.
