Overview

The combination of testosterone and thiotepa was historically utilized in the treatment of breast cancer, with its appearance in medical literature dating back several decades. Thiotepa functions as an alkylating agent, inducing irreparable DNA damage to prevent tumor cell division. Testosterone, on the other hand, is a hormone primarily indicated for treating hypogonadism. When used together, these medications exhibit complex bidirectional pharmacokinetic interactions: testosterone can increase the metabolism of thiotepa, while thiotepa can decrease the metabolism of testosterone. This combination therapy is primarily of historical interest and is not considered a standard treatment in modern clinical practice.
