Overview

TetMYB Vaccine is a DNA-based cancer vaccine developed to stimulate the immune system to recognize and attack tumor cells that overexpress the MYB transcription factor. The vaccine is constructed using a pVAX1 plasmid vector encoding a fusion protein consisting of universal tetanus toxin T-cell epitopes flanking an inactivated MYB gene. MYB is frequently overexpressed in colorectal cancer (CRC) and acts as a driver mutation in adenoid cystic carcinoma (AdCC), making it an attractive immunotherapeutic target. Preclinical studies demonstrated that vaccination with TetMYB induces tumor regression, establishes immune memory, and protects against tumor rechallenge. In clinical development, the vaccine has been tested both as monotherapy and in combination with anti-PD-1 antibody therapy (such as BGB-A317) for patients with advanced or metastatic solid cancers, including CRC and AdCC[2][5][9][10].
