Overview

Tetraarsenic oxide (As4O6), also known as TAO or KML001, is an inorganic arsenic compound investigated for its antineoplastic and anti-angiogenic properties. It is chemically distinct from the more common arsenic trioxide (As2O3) due to its cage-like dimeric structure. TAO demonstrates anti-cancer activity through multiple mechanisms, including the inhibition of the Akt signaling pathway in cancer cells and the downregulation of Matrix Metalloproteinase-2 (MMP-2) and Vascular Endothelial Growth Factor Receptor 2 (VEGFR2) in endothelial cells, which collectively suppress tumor growth and angiogenesis. Additionally, it has been shown to modulate autophagy in cancer cells. Developed primarily by Komipharm International, TAO has been evaluated in clinical trials for various malignancies, including prostate cancer and glioblastoma, and has shown preclinical efficacy in cervical cancer models.
