Overview

Tetrabromo-2-benzotriazole is a synthetic small molecule that acts as a potent and selective inhibitor of casein kinase II (CK2), also known as casein kinase II subunit alpha. The compound consists of a benzotriazole ring substituted with four bromine atoms at positions 4–7. It exhibits high thermal stability and resistance to degradation[1][3]. Tetrabromo-2-benzotriazole inhibits CK2 in an ATP/GTP-competitive manner by binding to the Val66 residue of the enzyme's catalytic subunit. This inhibition leads to downstream effects such as induction of caspase-dependent apoptosis and degradation of hematopoietic lineage cell-specific protein 1 in certain cell lines[4][5]. The compound is highly selective for CK2 over other kinases and has been widely used as a research tool in biochemical studies but is not approved for clinical use[3][5].
