Tetracaine is a long-lasting ester-type local anesthetic used primarily for surface anesthesia of the eyes, nose, throat, and mucous membranes. It is also applied to the skin before procedures such as intravenous cannulation to reduce pain. Tetracaine works by blocking voltage-gated sodium channels on nerve cell membranes, thereby inhibiting nerve impulse conduction and producing localized numbness. Its onset of action in the eye occurs within about 30 seconds and lasts less than 15 minutes when used topically. In clinical practice, it is considered much more potent than procaine and has a duration of action lasting up to several hours depending on the route of administration[1][2][6]. It may be used alone or in combination with other agents (e.g., lidocaine or adrenaline). In biomedical research, tetracaine can also act as an allosteric blocker of ryanodine receptors involved in calcium release from intracellular stores[2][8].
