Tetrahydrocannabivarin (THCV) is a naturally occurring cannabinoid found in the Cannabis sativa plant. It is a propyl analogue of tetrahydrocannabinol (Δ9-THC), differing by the length of its alkyl side chain, and is considered a major phytocannabinoid present in some cannabis strains. Unlike THC, THCV typically lacks psychoactive effects at standard doses and may even counteract some of THC’s intoxicating properties. Mechanistically, THCV acts as an antagonist at the Cannabinoid 1 receptor (CB1) and as a partial agonist at the Cannabinoid 2 receptor (CB2). It also exhibits agonist activity at GPR55 and l-alpha-lysophosphatidylinositol receptors, with additional evidence suggesting activation of 5HT1A serotonin receptors. Preclinical studies indicate that THCV can suppress appetite, improve glycemic control, enhance insulin sensitivity, reduce lipid accumulation, and provide neuroprotection. These properties make it a candidate for managing metabolic disorders such as obesity and type 2 diabetes; it has also been investigated for potential benefits in conditions like seizures and schizophrenia. However, purified or isolated THCV is not approved for any medical use nor available as a marketed pharmaceutical product[2][4][6][8].
