Tetraxetan, also known as DOTA (1,4,7,10-tetraazacyclododecane-1,4,7,10-tetraacetic acid), is a macrocyclic chelating agent produced as an active pharmaceutical ingredient (API) by Laboratorios Juste. It is a small molecule designed to sequester metal ions, such as gadolinium, gallium, and lutetium, within its 12-membered tetraaza ring and four acetic acid arms. This chelation process creates highly stable, non-toxic complexes suitable for medical use. Tetraxetan is a fundamental component in the production of gadoteric acid, a macrocyclic contrast agent used in magnetic resonance imaging (MRI). Additionally, it is a cornerstone of the radiopharmaceutical industry, where it acts as a bifunctional chelator conjugated to peptides or antibodies to deliver radionuclides for diagnostic PET/SPECT imaging or targeted radionuclide therapy in oncology.
