Tezepelumab + cat immunotherapy is a combination therapeutic approach evaluated for the induction of long-term immune tolerance in individuals with cat allergy. The regimen consists of **tezepelumab** (AMG 157), a human monoclonal antibody that targets and neutralizes **thymic stromal lymphopoietin (TSLP)**, and standardized **cat allergen immunotherapy** (allergy shots). TSLP is an epithelial-derived cytokine, or alarmin, that plays a critical role in initiating the allergic inflammatory cascade by activating dendritic cells and promoting type 2 immune responses. By blocking TSLP, tezepelumab aims to suppress the underlying inflammation while the concurrent administration of cat allergen extract desensitizes the immune system. This combination was primarily studied in the Phase 1/2 **CATNIP** (Cat Allergy Immunotherapy) trial, sponsored by the **National Institute of Allergy and Infectious Diseases (NIAID)** and conducted by the Immune Tolerance Network, to determine if it could provide faster and more durable symptom relief compared to allergy shots alone.
