Drug intelligence / Profile preview
TF2D-5a is an all-D-enantiomeric peptide ligand (21-mer) developed by Priavoid GmbH in collaboration with Forschungszentrum Jülich and Heinrich Heine University Düsseldorf. It is designed to target and disassemble toxic tau protein aggregates into harmless physiological tau monomers, acting via a thermodynamic mode of action. TF2D-5a is a derivative of TF2D-5, featuring five additional D-arginine residues at the C-terminus to enhance cell penetration. It has demonstrated nanomolar binding affinity to tau, inhibition of tau aggregation in vitro and in cell-based assays, and blood-brain barrier penetration. The compound is being developed as a potential disease-modifying treatment for Alzheimer's disease and other tauopathies.
