TFB-00640 is a potent, selective, orally administered small molecule antagonist of G protein-coupled receptor 35 (GPR35) being developed by ThirtyFiveBio for the treatment of ulcerative colitis. GPR35 is a genetically validated target in inflammatory bowel disease, and its activation is associated with epithelial dysfunction and damage in colitis. By inhibiting GPR35, TFB-00640 is designed to prevent harmful signaling in the gut lining, thereby protecting the gut epithelium from inflammatory damage and creating a protective 'gut epithelial shield.' The drug is currently in preclinical development, with investigational new drug (IND)-enabling studies underway to support the initiation of clinical trials.
