Overview

**TfRMAb-TNFR** is an investigational brain-penetrant recombinant fusion protein developed in academic research at Chapman University for neurodegenerative disease. It fuses a mouse transferrin receptor antibody transport domain to the extracellular domain of the type II human tumor necrosis factor alpha receptor, creating a soluble TNF-alpha decoy receptor that is transported across the blood-brain barrier through transferrin receptor-mediated transcytosis. In transgenic mouse models of Alzheimer’s disease and tauopathy, chronic dosing reduced neuroinflammatory and pathological markers, including microgliosis, amyloid-beta pathology, and tau phosphorylation, with evidence of improved memory-related outcomes in APP/PS1 mice.
