Overview

TG-1000 is a novel small molecule antiviral drug developed for the treatment of acute uncomplicated influenza A and B infections. It acts as a prodrug that is rapidly metabolized into its active form, TG-0527. The primary mechanism of action is inhibition of the influenza virus cap-dependent endonuclease (CEN), thereby blocking the cap-snatching process essential for viral mRNA transcription and replication. This mechanism allows it to be effective against both influenza A and B viruses, including avian H7N9 strains and neuraminidase-resistant variants. Clinical studies have shown that oral administration of TG-1000 leads to rapid onset of action, long-lasting viral suppression, good tolerability with a safety profile similar to placebo, and no significant food effect on pharmacokinetics. The drug has completed phase III trials in China with demonstrated efficacy in reducing symptom duration compared to placebo[1][4][5][6].
