Overview

TG-1031 is a recombinant prophylactic cancer vaccine developed by Transgene. It utilizes an attenuated vaccinia virus (Copenhagen strain) as a vector to deliver genetic sequences encoding human MUC1 and interleukin 2 (IL‑2). The vaccine is designed to stimulate the immune system by enhancing IL‑2 activity and promoting immune recognition of the tumor-associated antigen MUC1. Its primary indication was for immunotherapy in advanced or metastatic breast cancer. The mechanism involves stimulating both IL‑2 pathways and MUC1-specific immune responses to induce anti-tumor immunity. Clinical studies demonstrated immunogenicity with increases in T memory cells and cytotoxic T lymphocytes specific for MUC1; however, development has been discontinued after reaching Phase 2 trials[1][3][5].
