Overview

TG-4318 is a small molecule inhibitor of dipeptidyl peptidase 1 (DPP1), also known as cathepsin C, being developed by TaiGen Biotechnology. DPP1 is a lysosomal cysteine protease responsible for the activation of neutrophil serine proteases (NSPs), such as neutrophil elastase, cathepsin G, and proteinase 3, during neutrophil maturation in the bone marrow. In patients with non-cystic fibrosis bronchiectasis (NCFBE), excessive NSP activity in the lungs leads to chronic airway inflammation, mucus hypersecretion, and progressive lung damage. By inhibiting DPP1, TG-4318 aims to reduce the levels of active NSPs, thereby mitigating neutrophil-driven inflammation and preserving lung function in patients with NCFBE.
