TG01 + QS-21 Stimulon

TG01 + QS-21 Stimulon is a therapeutic cancer vaccine combination consisting of TG01, a polyvalent mixture of seven synthetic peptides representing common mutations in the KRAS and NRAS oncogenes, and QS-21 Stimulon, a saponin-based adjuvant. Developed by Circio (formerly Targovax) in collaboration with Agenus, the vaccine is designed to induce a robust T-cell mediated immune response against tumor cells harboring these specific RAS mutations, which are prevalent in over 90% of pancreatic cancers. The QS-21 Stimulon adjuvant enhances the immunogenicity of the peptides, aiming to overcome the immunosuppressive tumor microenvironment. The combination is currently being evaluated in Phase 1/2 clinical trials for the treatment of resected pancreatic cancer and multiple myeloma, both as a monotherapy and in combination with other agents such as checkpoint inhibitors.
