Tg1-90B is a novel small molecule radioligand developed for the detection and imaging of alpha-synuclein (α-syn) aggregates, which are the pathological hallmark of synucleinopathies such as Parkinson's disease (PD), dementia with Lewy bodies (DLB), and multiple system atrophy (MSA). It specifically targets "Site 2" on α-syn fibrils. Research conducted at the University of Pennsylvania has demonstrated that Tg1-90B acts as a "pan-α-syn" tracer, showing specific binding to α-syn inclusions in post-mortem brain tissue across PD, DLB, and MSA. This broad reactivity distinguishes it from other site-specific ligands that may only recognize aggregates in specific disease states. Tg1-90B is intended to serve as a diagnostic tool to improve the accuracy of diagnosis, monitor disease progression, and support the development of α-syn-targeting therapies.
