Overview

TG100948 is a small molecule multi-kinase inhibitor developed by TargeGen for the topical treatment of ophthalmic diseases, specifically targeting conditions characterized by vascular leakage and inflammation such as age-related macular degeneration (AMD) and diabetic macular edema (DME). The compound is designed to inhibit Src kinases and vascular endothelial growth factor (VEGF) receptors, which are key mediators of blood-retinal barrier breakdown. By stabilizing the vasculature of the eye through a topical application, TG100948 aimed to provide a less invasive alternative to intravitreal injections. Development of the compound was in the early preclinical or early clinical stages around 2007; however, following the acquisition of TargeGen by Sanofi in 2010, the ophthalmic program was not prioritized and is considered discontinued.
