TG1042 is an investigational gene therapy developed by Transgene, designed for the treatment of cutaneous lymphomas such as primary cutaneous B-cell lymphoma (PCBCL) and cutaneous T-cell lymphoma (CTCL). The therapy utilizes a non-replicating adenovirus serotype 5 (Ad5) vector to deliver the human interferon-gamma (IFN-gamma) gene directly into tumor lesions via intra-lesional injection. Once the vector transduces local cells, it triggers the localized production of IFN-gamma, a potent cytokine that induces anti-proliferative and pro-apoptotic effects on malignant B and T cells. Furthermore, TG1042 promotes an anti-tumor immune environment by upregulating MHC class I and II molecules, activating natural killer (NK) cells, and recruiting cytotoxic T-lymphocytes to the injection site. Clinical development reached Phase II, demonstrating efficacy in patients with relapsed or refractory cutaneous lymphomas.
