TG11-77 is a potent and selective small molecule antagonist of the prostaglandin E2 (PGE2) receptor subtype 2 (EP2). Developed by researchers at Emory University and the startup Thanakos, it is designed to mitigate the neuroinflammatory and neurodegenerative consequences of status epilepticus. Preclinical studies have shown that TG11-77 treatment can reduce microgliosis, alleviate post-seizure memory deficits, and significantly decrease the long-term accumulation of spontaneous recurrent seizure (SRS) burden. By blocking EP2 receptor signaling, the drug aims to interrupt the inflammatory cascades that contribute to the development of chronic epilepsy and associated cognitive impairments.
