TG4010 is an investigational **modified vaccinia Ankara viral-vector therapeutic cancer vaccine** developed by Transgene. The nonreplicating attenuated vaccinia vector encodes the full-length tumor-associated antigen **Mucin-1** and human **interleukin-2**, with the aim of eliciting Mucin-1-directed cellular antitumor immunity against Mucin-1-expressing tumors. It was principally evaluated by subcutaneous administration with chemotherapy, and later with nivolumab plus chemotherapy, in advanced non-small cell lung cancer. The randomized phase 2b TIME study reported improved progression-free survival in selected subgroups, particularly patients with low baseline triple-positive activated lymphocytes, but Transgene announced in December 2019 that it would stop further development of TG4010 following results from a phase 2 first-line chemoimmunotherapy study.
