Overview

TG6010 is an investigational **oncolytic vaccinia virus immunotherapy** from **Transgene** built on the company's **Invir.IO** platform. It is a genetically engineered Copenhagen-strain vaccinia virus with deletions in **thymidine kinase** and **ribonucleotide reductase** designed to favor replication in tumor cells, and it encodes **human cytidine deaminase**. The product is intended for cancer treatment as a biologic with multimodal activity: direct tumor-selective oncolysis, induction of immunogenic cell death, activation of innate and adaptive antitumor immunity, and local expression of human cytidine deaminase that depletes cytidine and deoxycytidine required for DNA replication, triggering DNA damage and apoptosis in cancer cells. Preclinical work also suggested a systemic antitumor effect through lowering available plasma cytidine, supporting development in solid tumors and potential combinations with agents targeting DNA replication and DNA repair.
