TG6050 is a clinical-stage oncolytic virus therapy developed using the Invir.IO platform. It is based on a genetically engineered vaccinia virus designed to selectively replicate within tumor cells and induce a strong immune response against tumors. The virus has been armed to express two therapeutic payloads within the tumor microenvironment: human interleukin-12 (IL-12), a cytokine that stimulates powerful antitumor immune responses, and a full-length anti-CTLA4 monoclonal antibody, which acts as an immune checkpoint inhibitor. By delivering these agents directly into tumors via viral infection, TG6050 aims to achieve high local concentrations for improved efficacy while minimizing systemic toxicity compared to conventional administration of IL-12 or anti-CTLA4 therapies. Preclinical studies have demonstrated robust antitumor activity in multiple murine models through both innate and adaptive immune mechanisms, with evidence of remodeling the tumor microenvironment from immunologically "cold" to "hot." The drug is administered intravenously and is currently being evaluated in Phase 1 clinical trials for metastatic non-small cell lung cancer.
