Overview

tgAAC09 is a recombinant adeno-associated virus serotype 2 (rAAV2) vector-based vaccine designed for the prophylaxis of HIV-1 infection. The vaccine construct contains a DNA genome with a cytomegalovirus (CMV) promoter driving the expression of a fusion protein comprising HIV-1 Clade C Gag, Protease (PR), and a truncated portion of the Reverse Transcriptase (RT) antigens. Developed by Targeted Genetics Corporation in collaboration with the International AIDS Vaccine Initiative (IAVI), tgAAC09 was manufactured using a specialized HeLa-based cell line (T3B12-5B) and an adenovirus helper. In clinical trials, the vaccine was evaluated for its ability to induce durable T-cell and B-cell immune responses, although development was ultimately discontinued following Phase 2 studies.
