Overview

TGFβR-KO CAR-EGFR T Cells are a form of chimeric antigen receptor (CAR) T cell therapy engineered to target the epidermal growth factor receptor (EGFR) on tumor cells while being resistant to immunosuppression mediated by transforming growth factor-beta (TGF-β). This resistance is achieved by knocking out the gene encoding the type II TGF-β receptor (TGFBR2) in the CAR-T cells using CRISPR-Cas9 genome-editing technology. The dual modification allows these engineered cells to maintain anti-tumor activity and proliferation even within highly immunosuppressive tumor microenvironments, such as those found in solid tumors. The primary indication under investigation is advanced unresectable or metastatic biliary tract cancer, with ongoing clinical trials evaluating safety and efficacy[3][1][2].
