Drug intelligence / Profile preview
TGF-β blocking antibodies are monoclonal antibodies designed to bind and neutralize transforming growth factor-beta (TGF-β) ligands (such as TGF-β1, TGF-β2, and/or TGF-β3). TGF-β is a multifunctional cytokine that plays a critical role in regulating cell growth, differentiation, immune responses, and tissue remodeling. In pathological states, dysregulated TGF-β signaling is implicated in tissue fibrosis, tumor immune evasion, and bone remodeling disorders. In bone biology, TGF-β signaling regulates the quiescence and reactivation of osteoblasts; blocking TGF-β has been shown to stimulate the transition of quiescent bone lining cells into active, bone-forming osteoblasts. Consequently, TGF-β blocking antibodies are being investigated, often in combination with anabolic agents like anti-sclerostin antibodies, as potential therapeutic strategies to enhance bone mass and treat severe osteoporosis.
