A therapeutic peptide vaccine designed to prevent aortic aneurysm formation, particularly in the context of Marfan syndrome (MFS). The vaccine targets Transforming Growth Factor-β1 (TGF-β1) by utilizing specific peptide sequences (p.304-313 and p.313-322) common to both humans and mice as antigens. These antigens are conjugated to a carrier protein and administered with an adjuvant to induce the production of neutralizing antibodies against TGF-β1. By neutralizing TGF-β1, the vaccine aims to attenuate the overactive TGF-β signaling pathway that contributes to aortic wall thickening, elastic fiber degeneration, and proteoglycan deposition. Research conducted by the University of Tokyo and Osaka University has demonstrated its efficacy in suppressing aortic diameter enlargement in mouse models of MFS.
