A TGF-β1 neutralizing antibody is a biologic agent designed to bind and inhibit the activity of Transforming Growth Factor-beta 1 (TGF-β1), a pleiotropic cytokine that plays a critical role in cancer progression. While TGF-β1 acts as a tumor suppressor in early-stage disease, it often switches to a tumor-promoting role in advanced cancers, driving epithelial-to-mesenchymal transition (EMT), metastasis, and immune suppression. In pancreatic ductal adenocarcinoma (PDA), particularly in tumors overexpressing tumor-associated Mucin-1 (tMUC1), TGF-β1 signaling is redirected through non-canonical pathways (such as Erk1/2) to promote invasion and resistance to apoptosis. Neutralizing antibodies targeting TGF-β1 aim to block these pathways, thereby reducing tumor growth and enhancing the therapeutic response, especially in tMUC1-high patient populations. These antibodies are being explored as potential therapeutic agents to reverse TGF-β-mediated immune evasion and tumor progression.
