Overview

TGF-β1 siRNA is a small interfering RNA (siRNA) molecule specifically designed to silence the gene expression of transforming growth factor-beta 1 (TGF-β1). TGF-β1 is a cytokine implicated in a variety of fibrotic, oncologic, and immunological diseases due to its role in promoting immune suppression, tumor angiogenesis, tissue fibrosis, and scar formation. TGF-β1 siRNA acts by binding to TGF-β1 mRNA and promoting its degradation, thereby preventing TGF-β1 protein translation. This therapeutic approach has been investigated in several formulations and delivery modalities, including synthetic siRNA, nanoparticle delivery, and exosome-based delivery, and is explored as both a monotherapy and as part of dual-targeting approaches (e.g., combined with COX-2 siRNA). The intended therapeutic indications include cancer (to enhance immunotherapy and inhibit tumor progression), liver fibrosis, idiopathic pulmonary fibrosis, hypertrophic scar, and cardiac repair[1][2][3][5][6][7][8][9][10].
