TGF-beta1 neutralizing antibody refers to a class of monoclonal antibodies designed to selectively bind and neutralize transforming growth factor beta-1 (TGF-beta1), a pleiotropic cytokine that plays a critical role in tissue fibrosis, immune suppression, and tumor progression. By blocking TGF-beta1, these antibodies aim to prevent downstream SMAD2/3 signaling, thereby inhibiting the activation of myofibroblasts, reducing extracellular matrix (ECM) deposition, and reversing immunosuppression in the tumor microenvironment. Several clinical-stage candidates targeting this pathway have been developed, including metelimumab (CAT-192) and LY2382770, which have been investigated for fibrotic conditions such as systemic sclerosis and diabetic nephropathy, though many have faced challenges in clinical trials due to the complex, context-dependent biology of TGF-beta1. More recently, candidates like nisevokitug (NIS793) and RG6440 (RO7496353) have been evaluated in oncology settings to modulate the tumor microenvironment.
