TGFBR2-KO CD70-CAR NK cells is an investigational allogeneic chimeric antigen receptor (CAR) natural killer (NK) cell therapy developed by the M.D. Anderson Cancer Center. Derived from umbilical cord blood, these NK cells are genetically engineered to express a CAR targeting CD70, a tumor-associated antigen highly expressed in clear cell renal cell carcinoma (ccRCC), mesothelioma, and osteosarcoma. To overcome the immunosuppressive tumor microenvironment, the cells undergo CRISPR/Cas9-mediated knockout of the transforming growth factor-beta receptor type 2 (TGFBR2) and the cytokine-inducible SH2-containing protein (CISH), which are negative regulators of NK cell function. The therapy is further enhanced by the ectopic expression of Interleukin-15 (IL-15) to improve cell persistence and proliferation, and includes an inducible caspase 9 (iC9) safety switch for controlled elimination of the cells if toxicity occurs.
