TGI-17b is a potential first-in-class trispecific antibody designed to simultaneously target Programmed Cell Death Protein 1 (PD-1), Vascular Endothelial Growth Factor (VEGF), and Interleukin-2 (IL-2) with an alpha-bias. By integrating immune checkpoint inhibition, anti-angiogenesis, and biased cytokine signaling into a single molecule, TGI-17b aims to overcome resistance to standard immune checkpoint inhibitors and convert immunologically "cold" tumors into "hot" tumors. The IL-2 component is engineered with an alpha-bias (high affinity for IL-2Rα/CD25), which selectively directs the cytokine's stimulatory signals toward activated effector T cells while minimizing the systemic toxicity and regulatory T cell activation often associated with non-biased IL-2 therapies. Preclinical data presented at AACR 2026 demonstrated that TGI-17b has superior anti-tumor efficacy compared to PD-1/VEGF or PD-1/IL-2 bispecific antibodies and maintains a favorable safety profile, positioning it as a potential cornerstone for pan-cancer immunotherapy.
