TGIF1-silenced adipose-derived stem cell exosomes are an experimental exosome-based therapy developed by researchers at Shanghai Tenth People's Hospital. These exosomes are harvested from adipose-derived mesenchymal stem cells (ADSCs) in which the TGFB-induced factor homeobox 1 (TGIF1) gene has been silenced, typically using RNA interference techniques such as shRNA. TGIF1 is a transcriptional corepressor that negatively regulates the TGF-beta/Smad signaling pathway. By silencing TGIF1 in the parent stem cells, the resulting exosomes are enriched with a specific cargo of microRNAs and proteins that enhance tenogenic differentiation and collagen synthesis while potentially reducing peritendinous adhesion and fibrosis. This therapy is primarily investigated for the treatment of musculoskeletal conditions, including rotator cuff injuries, cruciate ligament tears, and various forms of tendinitis, with the goal of improving the quality of tendon-to-bone healing and functional recovery.
