TGM-291 is a GalNAc-conjugated small interfering RNA (siRNA) therapeutic candidate developed by Tangram Therapeutics (formerly E-Therapeutics) for the treatment of cardiometabolic diseases, including Type 2 diabetes, cardiovascular disease, and metabolic syndrome. Utilizing the company's proprietary GalOmic technology platform, the drug is designed to mediate RNA interference (RNAi) to silence a specific, undisclosed gene in liver hepatocytes that has strong human genetic validation. Preclinical studies have demonstrated that TGM-291 provides pleiotropic benefits across multiple cardiovascular risk factors, including improvements in insulin sensitivity, cholesterol levels, and fibrinogen. It is positioned as a potential treatment for residual cardiovascular risk that persists after standard LDL-cholesterol lowering therapies. TGM-291 is currently in the preclinical stage of development.
