TGM-312

TGM-312 (also known as TGM-312-SC01) is a long-acting, engineered fibroblast growth factor 21 (FGF21) analog developed by Tangram Therapeutics for the treatment of metabolic dysfunction-associated steatohepatitis (MASH). Utilizing the company's Tangram-Engineered Multi-specifics (TEM) platform, TGM-312 is designed to overcome the short half-life and poor stability of native FGF21 while maintaining high potency and selectivity. FGF21 is a key metabolic regulator that improves insulin sensitivity, reduces hepatic lipid accumulation, and exerts anti-inflammatory and anti-fibrotic effects by signaling through the FGFR1c/beta-Klotho receptor complex. TGM-312 is currently in Phase 1/2 clinical development, where it is being evaluated for its ability to improve liver health and metabolic parameters in patients with MASH.
