Overview

TGN-S11 is a small molecule drug developed by Toragen for the treatment of human papillomavirus (HPV)-associated cancers and infections. Originally approved as an antimicrobial agent, TGN-S11’s mechanism of action involves inhibition of the HPV E5 oncogene protein, which is implicated in the pathogenesis of HPV-related malignancies. The drug has demonstrated both antiviral and antineoplastic activities, with evidence showing it decreases growth of HPV-positive tumor cells and improves survival in preclinical models. Currently, TGN-S11 is being evaluated in a Phase 1 clinical trial as monotherapy and in combination with Keytruda (pembrolizumab) for patients with relapsed, resistant, or metastatic HPV-associated cancers. The trial has shown promising safety results and preliminary signs of efficacy including reductions in tumor size and circulating tumor HPV DNA.
