TGRX-814 is an orally bioavailable, highly selective small molecule inhibitor of the anti-apoptotic protein B-cell lymphoma 2 (BCL2), developed for the treatment of hematological malignancies. By binding to and inhibiting BCL2, TGRX-814 restores apoptotic processes in tumor cells, leading to pro-apoptotic and antineoplastic effects. It demonstrates high selectivity for BCL2 over BCL-XL and has been optimized for improved oral metabolism, increased in vivo exposure, and reduced clearance compared to existing drugs such as venetoclax. TGRX-814 is currently being evaluated in phase I/II clinical trials as both monotherapy and combination therapy for various blood cancers including non-Hodgkin lymphoma (NHL), acute myeloid leukemia (AML), acute lymphoblastic leukemia (ALL), myelodysplastic syndromes (MDS), chronic lymphocytic leukemia (CLL) with or without del(17p)/TP53 mutation, small lymphocytic lymphoma (SLL), diffuse large B-cell lymphoma (DLBL), and multiple myeloma (MM)[1][2][3][4][5].
