TGW101

TGW101 is an antibody-drug conjugate (ADC) developed by Tagworks Pharmaceuticals. It targets tumor-associated glycoprotein 72 (TAG72), a clinically validated non-internalizing antigen widely expressed in solid tumors, including breast, colorectal, stomach, lung, pancreatic, prostate, and ovarian cancers. Unlike conventional ADCs that require internalizing targets for efficacy, TGW101 leverages Tagworks' proprietary Click-to-Release technology to enable controlled drug release in the tumor microenvironment. The ADC consists of a TAG72-binding diabody conjugated with TCO-linked monomethyl auristatin E (MMAE), a potent tubulin inhibitor with a well-documented bystander effect. In therapy administration, the ADC is first allowed to bind and clear from circulation; then a trigger molecule is injected systemically to cleave the tumor-bound ADC and release MMAE locally into surrounding tumor cells[1][3][4][6]. This approach aims to expand the therapeutic window of ADCs and address tumors with heterogeneous or low target expression.
