TGX-007 is an investigational **AAV1-mediated dual-payload gene therapy** developed by Trogenix for newly diagnosed and recurrent glioblastoma. The vector uses a tumour-cell-selective synthetic super-enhancer designed to be active in glioblastoma stem-like cell states and encodes **herpes simplex virus thymidine kinase** and **interleukin-12**. HSV thymidine kinase is intended to convert subsequently administered valaciclovir into a cytotoxic metabolite within transduced tumour cells, while interleukin-12 is intended to stimulate antitumour immune activity. TGX-007 is administered directly into tumour tissue by MRI-guided convection-enhanced delivery and is being evaluated in the Phase I/II ADePT study.
