TH-1940 is an investigational peptide-drug conjugate (PDC) developed by Theratechnologies, designed to target the Sortilin-1 (SORT1) receptor. SORT1 is frequently overexpressed in various aggressive cancers, including ovarian, triple-negative breast, and colorectal cancers, and plays a role in protein trafficking and internalisation. TH-1940 consists of a proprietary SORT1-targeting peptide (TH19P01) conjugated to a potent topoisomerase I inhibitor payload, such as exatecan. Upon binding to SORT1, the conjugate is internalised via endocytosis, where the cytotoxic payload is released to induce DNA strand breaks and cell death. This targeted delivery mechanism is intended to enhance the therapeutic efficacy of the topoisomerase inhibitor while reducing systemic side effects compared to traditional chemotherapy. TH-1940 is currently in preclinical development as part of Theratechnologies' oncology platform.
