**TH-407b** is a potent and selective small molecule inhibitor of Toll-like receptor 7 (TLR7). It dose-dependently inhibits R848-induced TLR7 activation with an IC50 of 0.23 µM in HEK-Blue hTLR7 cells[1]. This molecule stabilizes TLR7 in its resting/inactivated state, thereby suppressing TLR7-mediated pro-inflammatory signaling. TH-407b demonstrates anti-inflammatory activity in cell cultures and peripheral blood mononuclear cells from systemic lupus erythematosus (SLE) patients, and shows efficacy in mouse models of SLE[4]. It is being developed by Toll Biotech Co.Ltd.(Beijing) for immune system diseases, with systemic lupus erythematosus as its primary indication[4]. The drug is in preclinical development[4].
