Drug intelligence / Profile preview
TH10785 is a first-generation small molecule organocatalytic switch (ORCA), also referred to as an "amplizyme," that acts as an activator of 8-oxoguanine DNA glycosylase 1 (OGG1). Rather than acting as a conventional inhibitor, TH10785 binds to the active site of OGG1 and enhances its catalytic activity 10-fold, introducing a de novo β,δ-lyase enzymatic function that bypasses the need for apurinic endonuclease 1 (APE1) and redirects base excision repair of oxidative DNA damage to a polynucleotide kinase phosphatase (PNKP1)-dependent pathway. Developed by researchers at Karolinska Institutet and SciLifeLab, TH10785 is being investigated preclinically for its potential therapeutic applications in progressive fibrotic interstitial lung disease (ILD), idiopathic pulmonary fibrosis (IPF), cancer, Alzheimer's disease, and age-related conditions.
