**TH1904** is a peptide-drug conjugate (PDC) developed by Theratechnologies, consisting of the SORT1-targeting peptide TH19P01 conjugated to two molecules of doxorubicin via a 3,3’-dimethylglutarate linker. It selectively internalizes into SORT1-expressing cancer cells, enabling precise delivery of the cytotoxic payload to inhibit tumor growth and vasculogenic mimicry (VM), a chemotherapy-resistant process associated with poor prognosis in ovarian and triple-negative breast cancers (TNBC). Preclinical data demonstrate high tumor accumulation, low off-target effects in healthy tissues, and superior efficacy over free doxorubicin or liposomal formulations (Doxil) in ES-2 ovarian cancer and MDA-MB-231 TNBC models, where it disrupts VM at low nanomolar concentrations.
