Thailanstatin D is a potent cytotoxic natural product belonging to the thailanstatin family, originally isolated from the bacterium *Burkholderia thailandensis*. It functions as a spliceosome inhibitor by specifically targeting the SF3b (Splicing Factor 3b) complex, which is essential for the recognition of the branch point sequence during pre-mRNA splicing. By disrupting the splicing process, thailanstatin D induces cell cycle arrest and apoptosis in cancer cells. Due to its high potency and distinct mechanism of action compared to traditional tubulin or DNA-targeting agents, it is primarily utilized as a payload for antibody-drug conjugates (ADCs).
