Overview

THB001 is a first-in-class, oral small-molecule inhibitor of the KIT receptor tyrosine kinase, developed by Third Harmonic Bio. KIT is a cell surface receptor that acts as the master regulator of mast cell function and survival. By binding to the tyrosine kinase domain of KIT, THB001 prevents autophosphorylation and dimerization of the receptor, thereby blocking downstream signaling and leading to depletion of mast cells. This mechanism targets mast cell-driven inflammation implicated in allergic and inflammatory diseases affecting skin, airways, and gastrointestinal tract. In preclinical studies, THB001 demonstrated potent activity with high selectivity for KIT over other kinases and robust pharmacodynamic effects in early clinical trials as measured by serum tryptase levels. Clinical development was focused on chronic inducible urticaria; however, Phase 1b studies were discontinued due to asymptomatic elevations in liver enzymes (transaminitis) linked to a reactive metabolite pathway not predicted by earlier toxicology studies[1][4][6][8].
